The BrainPortal Platform

ChainGen Bio is a clinical-stage biotechnology company developing brain-penetrant antibodies and oligonucleotide therapeutics for central nervous system diseases, powered by our proprietary AI-designed BrainPortal Platform for blood-brain barrier transport. The platform is engineered to combine precision targeting, scalable manufacturability, and translational efficiency across both antibody and oligo-based modalities.

AI-Powered Design

De novo designed, self-folding TfR1-binding peptides engineered for receptor-selective, luminal-to-abluminal transport across the BBB.

Plug-and-Play

Composed entirely of natural amino acids, with scalable fermentation and strong stability. Compatible with both antibodies (Plug-and-Play) and siRNA payloads (ARC).

Small is “Big“

Among the smallest BBB shuttles at approximately 80 amino acids, supported by wholly owned IP and a demonstrated path from design to first-in-human evaluation.

MECHANISM

Crossing the Blood-Brain Barrier via TfR1

The blood-brain barrier (BBB) is formed by tightly connected brain vascular endothelial cells that protect the central nervous system, but also limit therapeutic access to sites of disease. Transferrin receptor 1 (TfR1) functions as a natural iron-transport gateway at the BBB. Our BrainPortal constructs bind TfR1 on the brain endothelium and are transported across the barrier into brain tissue through receptor-mediated transcytosis, without relying on delivery through the choroid plexus or cerebrospinal fluid.

Mechanism of receptor-mediated transcytosis across the BBB via TfR1

BrainPortal Plus (CD98HC) expands the platform beyond TfR1, with additional transporter programs targeting INSR and IGF1R currently in development.

Our Pipeline

A clinical-stage pipeline of brain-penetrant programs targeting amyloid-beta, CD20, alpha-synuclein, and undisclosed targets for Alzheimer's disease, multiple sclerosis, Parkinson's disease, and Huntington's disease.
